    The Aging Imperative in Maine: Present Realities and Future Prospects

    Maine has one of the oldest populations in the United States. In this article, Helen Miltiades and Lenard Kaye, guest editor for this special issue, provide an overview of Maine’s aging population and the challenges—and opportunities—faced in the “revolution in aging” that is taking place. They point out how the growing older adult population is expected to place greater demands on family caregivers, on the health and long-term care systems, and on state and federal budgetary and policy decisions. Describing some of Maine’s innovative public and private sector responses, they note that Maine has been in the forefront in providing home and community-based care for its frailer elders. At the same time, they emphasize that the aging population presents expanded opportunities, as increasing numbers of healthier older adults choose to remain in the labor force, to participate in education, and to play active roles in philanthropy, the arts, and as volunteers

The Universal Cloud and Aerosol Sounding System (UCASS) is an optical particle counter (OPC), which uses wide-angle elastic light scattering for the high-precision sizing of fluid-borne particulates. The UCASS has up to 16 configurable size bins, capable of sizing particles in the range 0.4–40 µm in diameter. Unlike traditional particle counters, the UCASS is an open-geometry system that relies on an external air flow. Therefore, the instrument is suited for use as part of a dropsonde, balloon-borne sounding system, as part of an unmanned aerial vehicle (UAV), or on any measurement platform with a known air flow. Data can be logged autonomously using an on-board SD card, or the device can be interfaced with commercially available meteorological sondes to transmit data in real time. The device has been deployed on various research platforms to take measurements of both droplets and dry aerosol particles. Comparative results with co-located instrumentation in both laboratory and field settings show good agreement for the sizing and counting ability of the UCASS.Peer reviewe

    Construction and properties of a mutant of herpes simplex virus type 1 with glycoprotein H coding sequences deleted

    A mutant of herpes simplex virus type 1 (HSV-1) in which glycoprotein H (gH) coding sequences were deleted and replaced by the Escherichia coli lacZ gene under the control of the human cytomegalovirus IE-1 gene promoter was constructed. The mutant was propagated in Vero cells which contained multiple copies of the HSV-1 gH gene under the control of the HSV-1 gD promoter and which therefore provide gH in trans following HSV-1 infection. Phenotypically gH-negative virions were obtained by a single growth cycle in Vero cells. These virions were noninfectious, as judged by plaque assay and by expression of I-galactosidase following high-multiplicity infection, but partial recovery of infectivity was achieved by using the fusogenic agent polyethylene glycol. Adsorption of gH-negative virions to cells blocked the adsorption of superinfecting wild-type virus, a result in contrast to that obtained with gD-negative virions (D. C. Johnson and M. W. Ligas, J. Virol. 62:4605-4612, 1988). The simplest conclusion is that gH is required for membrane fusion but not for receptor binding, a conclusion consistent with the conservation of gH in all herpesviruses

    Personality profiles and persuasion: An exploratory study investigating the role of the Big-5, Type D personality and the Dark Triad on susceptibility to persuasion

    The present study investigated the relationship between personality profiles and susceptibility to persuasion.
 Participants (N = 316) were recruited for an online questionnaire and asked to complete self-reported measures of their personality – Big-5, Dark Triad and Type D. Individual differences in susceptibility to persuasion were also explored using Cialdini's model of persuasion. Latent profile analysis identified three distinct profiles which were labelled Socially Apt, Fearful and Malevolent. These profiles were correlated with scores on the persuasion sub-scales – authority, commitment, liking, scarcity, reciprocity, consensus – and a number of interesting associations were identified. The malevolent profile self-reported as more susceptible to a higher use of scarcity relative to the other principles of persuasion, and was least susceptible to reciprocity and authority. The socially apt profile appear to be more inclined to be persuaded to do something if it is consistent with their beliefs or a prior act whereas individuals in the Fearful profile were more likely to report obeying those in authority and going along with a crowd. Implications for persuasion are discussed along with research on personality types

    How emotional are emoji?: Exploring the effect of emotional valence on the processing of emoji stimuli.

    Emoji are vastly becoming an integral part of everyday communication, yet little is understood about the extent to which these are processed emotionally. Previous research shows that there is a processing advantage for emotionally-valenced words over neutral ones, therefore if emoji are indeed emotional, one could expect an equivalent processing advantage. In the Pilot Study, participants (N = 44) completed a lexical decision task to explore accuracy and response latency of word, face and emoji stimuli. This stimuli varied in emotional valence (positive vs. neutral). Main effects were found for stimuli type and valence on both accuracy and latency, although the interaction for accuracy was not significant. That is, there were processing advantages of positively-valenced stimuli over neutral ones, across all stimuli types. Also, faces and emoji were processed significantly more quickly than words, and latencies between face and emoji stimuli, irrespective of valence were largely equivalent. The Main Study recruited 33 participants to undertake a modified and extended version of the lexical decision task, which included three valence conditions (positive, negative and neutral) per stimuli type. Although no main effects were found for accuracy, there was a significant main effect found for stimuli but not for valence on latency. Namely, that word stimuli irrespective of valence were processed significantly more slowly than face or emoji stimuli. There was not a significant interaction between stimuli and valence, however. Therefore, overall although there was partial support for a processing advantage of emoji stimuli, this was not replicated across the studies reported here, suggesting additional work may be needed to corroborate further evidenc

    VALIDATE:Exploiting the synergy between complex intracellular pathogens to expedite vaccine research and development for tuberculosis, leishmaniasis, melioidosis and leprosy

    For several complex intracellular pathogens, we have an urgent need for effective vaccines and yet there are common barriers to vaccine development. These diseases, including tuberculosis, leishmaniasis, leprosy and melioidosis, cause a huge burden of disease and disproportionately affect low and middle income countries. They are therefore often neglected due to the marginalisation of affected populations and the poor predicted commercial return on investment. Barriers to vaccine development include an incomplete understanding of protective immunity and translation from the bench into clinical vaccine trials. The current linear approach to vaccine research and development for these pathogens, which involves basic research, vaccine design, and vaccine evaluation in preclinical challenge models and clinical trials, is inefficient for these complex intracellular pathogens. We have established a Global Challenges Research Fund Network for VAccine deveLopment for complex Intracellular neglecteD pAThogEns, “VALIDATE”, where we aim to adopt a more flexible, integrated cross-pathogen approach to accelerate vaccine research and clinical development for these four pathogens, by cross-pathogen analyses, cross-discipline collaborations, and repeated integration of data from human and animal studies. This network provides a unique opportunity to bring together individuals working on four exemplar complex intracellular neglected pathogens (M.tb, Leishmania spp., B. pseudomallei and M.leprae), which share a common lifestyle as pathogens of macrophages, induce similar end-stage pathologies and alter host immune and metabolic responses. The horizontal collaborations established throughout this network, together with the provision of a protected environment for early data sharing, will exploit these biological synergies. By interrogating mechanisms that lead from infection to disease, we will be able to develop common vaccine development strategies for these and other complex intracellular pathogens. Keyword
